Dr. Grams is the lead author of a study published that month in the journal JAMA Internal Medicine, where he and his team found that people who use Nexium or another PPI may raise their risk of kidney disease by up to 50%. These researchers also found that the risk of kidney disease goes up the longer a person uses a PPI, and that the risk is also increased in people who take two doses of these drugs a day. Later in 2016, another study which was published in the Journal of the American Society of Nephrology suggested that patients who take a PPI like Nexium are at a much higher risk of kidney disease and kidney failure than people who use a different class of acid medications. In fact, Nexium users may be up to 96% more likely to suffer end-stage kidney failure.

Kidney disease or kidney failure are not the only serious side effects linked to Nexium. This “purple pill” has also been associated with a higher risk of interstitial nephritis, which was the subject of an FDA warning in 2014. This condition is often a precursor to severe kidney injury or even kidney failure. The higher-than-average rate of interstitial nephritis in Nexium users may also contribute to the high rate of acute kidney injury in users of this and other PPIs. In 2015, research published in the journal CMAJ Open found that people who use a PPI like Nexium are at a 2.5 times higher risk for kidney disease.
